1 - To modify/add/remove panes you have to get to know how to build skins: This may give you some of the basics: www.themeswiki.org/Creating_your_first_DotNetNuke_Skin

2 - Yes, multiple "interfaces" can exist. The correct terminology in DotNetNuke is "skins

3 - Yes you can shift them by going to the page setting and selecting a new skin. Maybe another video that would be useful to you is this one:
